Effect of diets containing different levels of highly unsaturated fatty acids on physiological and immune responses in Pacific whiteleg shrimp Litopenaeus vannamei (Boone) exposed to handling stress ArchiMer
Mercier, Laurence; Racotta, Ilie; Yepiz-plascencia, Gloria; Muhlia-almazan, Adriana; Civera, Roberto; Quinones-arreola, Marcos F.; Wille, Mathieu; Sorgeloos, Patrick; Palacios, Elena.
Juveniles fed a diet containing a low or a high level of highly unsaturated fatty acids (HUFA) for 38 days were exposed to handling stress. In a first experiment, stress was applied daily for 30 days, after which the physiological and immunological variables were measured, whereas in a second experiment, stress was applied once and samples were obtained 1 and 24 h after the stressor event. Shrimp that were stressed for 30 days showed significantly lower survival, final weight and feed consumption compared with unstressed shrimp. The concentration of the high-density lipoprotein beta-glucan-binding protein was significantly higher in shrimp fed the high-HUFA diet. The glucose concentration in the haemolymph was significantly higher in long-term stressed...

Effect of unilateral and bilateral eyestalk ablation in Litopenaeus vannamei male and female on several metabolic and immunologic variables ArchiMer
Sainz Hernandez, J; Racotta, Ilie; Dumas, S; Hernandez Lopez, J.
Eyestalk ablation is the most common procedure to induce gonadic maturation in commercial hatcheries of penaeid shrimp. In addition to reproduction, other physiological and metabolic processes are affected by removal of the X-organ sinus gland complex located in the eyestalk. In this study, the effect of unilateral and bilateral eyestalk ablation on the concentration of several hemolymph metabolites and phenoloxidase system in female and male shrimp was investigated. As a consequence of reducing or suppressing molt-inhibiting hormone (MIH) production, the duration of the molting cycle was significantly shorter in eyestalk-ablated shrimp: bilaterally (10 days), unilaterally (17 days), and shrimp that were not ablated (24 days). Mortality was significantly...

Laboratory conditioning modifies properties of gills mitochondria from the Pacific oyster Crassostrea gigas ArchiMer
Dudognon, Tony; Guderley, Helga; Quere, Claudie; Soudant, Philippe; Racotta, Ilie; Kraffe, Edouard.
Although laboratory experiments allow greater control of environmental conditions than field studies, they have several drawbacks. To analyze physiological responses to forcing environmental variables, experimental conditions should mimic natural conditions as closely as possible. For filter-feeding organisms in particular, diet quality and quantity is one of the environmental parameters that can differ markedly between experimental and field conditions. In the hatchery, Pacific oysters, Crassostrea gigas, commonly show good physiological performance and growth on a mixed algal diet of Tisochrysis lutea, formerly Isochrysis aff. galbana clone Tahiti (T-Iso), and Chaetoceros calcitrans, presumably as it provides a good supply of essential polyunsaturated...

Reproductive performance, biochemical composition and fatty acid profile of wild-caught and 2nd generation domesticated Farfantepenaeus duorarum (Burkenroad, 1939) broodstock ArchiMer
Emerenciano, Mauricio; Cuzon, Gerard; Mascaro, Maite; Arevalo, Miguel; Norena-barroso, Elsa; Jeronimo, Gilberto; Racotta, Ilie; Gaxiola, Gabriela.
A 30-day trial was performed to evaluate the reproductive performance of wild and 2nd generation domesticated Farfantepenaeus duorarum broodstock. Changes in biochemical composition and fatty acids (FA) profile in the 1st and 4th spawn order females were used as indicators of nutritional condition. Wild population of F. duorarum presented significantly better reproductive outcomes as compared to domesticated ones. Wild spawners achieved significantly higher number of eggs per spawn, number of egg per spawn per g of spawner's body weight and number of nauplii per spawn (P<0.05). Latency period was significantly shorter (P<0.05) for wild than for domesticated females (11 vs 17 days). Wild spawners also presented less mortality (6 vs 12%), higher number...

The influence of dietary supplementation of arachidonic acid on prostaglandin production and oxidative stress in the Pacific oyster Crassostrea gigas ArchiMer
Seguineau, Catherine; Racotta, Ilie; Palacios, Elena; Delaporte, Maryse; Moal, Jeanne; Soudant, Philippe.
In a previous study, dietary supplementation with arachidonic acid (ARA) to oysters Crassostrea gigas increased haemocyte numbers, phagocytosis, and production of reactive oxygen species level (ROS) by haemocytes (Delaporte et al., 2006). To assess if the observed stimulation of these cellular responses resulted from changes of ARA-related prostaglandin (PG) production, we analysed prostaglandin E2 metabolite (PGEM) content on the same oysters fed three levels of ARA. Dietary supply of polyunsaturated fatty acids (PUFA) could also induce an oxidative stress that could similarly increase cellular responses; therefore, two indicators of oxidative stress were analysed: peroxidation level and antioxidant defence status. Together the observed positive...
